I'm looking for a classic novel or other Great-Books-type literature that contemplates the meaning of friendship. I don't know why, I'm just kind of in the mood for that. Anyone have any suggestions?

I have a couple Great Books for you:
De Amicitia (On Friendship) "A friend is a second self..."
Plato's Symposium
As for literature, Othello fits the bill well as a via negativa - Iago and Othello's case is in many ways more interesting than the Moor's and Desdemona's.
The theme of friendship can also be parsed out from the Englishness of it all, too, in Brideshead Revisited.
Also, the Iliad shows a great deal about friendship in the dynamic between Akilles and Patrokolus.
Also, a Tale of Two Cities.
Huck Finn (Huck and Jim)
I'd also round it out with, although I've never read it, The Four Feathers, which was made into several movies. Not as heavy weight as the rest, but perhaps a good study, if the movies do it any justice, just the same.
